Chlordiazepoxide is a benzodiazepine derivative used to treat severe anxiety, insomnia, anesthesia premedication, adjunct in management of seizures, muscle spasms in severe spastic disorders, alcohol withdrawal syndrome, sedation in minor surgical and medical procedures.
CNS drugs- (Anxiolytics)
Chlordiazepoxide is prescribed for the treatment of:
• Severe anxiety
• Insomnia
• Anesthesia premedication
• Adjunct in management of seizures
• Muscle spasms in severe spastic disorders
• Alcohol withdrawal syndrome
• Sedation in minor surgical and medical procedures
As advised by the physician.
According to the dosage forms the following routes of administration are available:
• Oral
• Parenteral
Chlordiazepoxide has anti-anxiety, sedative, appetite-stimulating and weak analgesic actions. The drug seems to block EEG arousal from stimulation in the brain stem reticular formation. The drug has been studied extensively in many species of animals and these studies are suggestive of action on the limbic system of the brain, which recent evidence indicates is involved in emotional responses. Hostile monkeys were made tame by oral drug doses which did not cause sedation. Chlordiazepoxide revealed a taming action with the elimination of fear and aggression. The taming effect of chlordiazepoxide was further demonstrated in rats made vicious by lesions in the septal area of the brain. The drug dosage which effectively blocked the vicious reaction was well below the dose which caused sedation in these animals.

Amitriptyline is metabolized to nortriptyline which inhibits the reuptake of norepinephrine and serotonin almost equally. Amitriptyline inhibits the membrane pump mechanism responsible for uptake of norepinephrine and serotonin in adrenergic and serotonergic neurons. Pharmacologically this action may potentiate or prolong neuronal activity since reuptake of these biogenic amines is important physiologically in terminating transmitting activity. This interference with the reuptake of norepinephrine and/or serotonin is believed by some to underlie the antidepressant activity of amitriptyline.
